Another set up question

OK,
I an going to take a 4 str, and set it up for Drop C. I will be using 65-125 strings. After I alter the nut, what is the first step I should take. Should I just string it up, tune it and let is sit overnight to see what the neck does, ot should I adjust the relief before I start?...

I always put the strings on and let it sit for awhile before making any trussrod adjustments... Everything may be alright to begin with so you may not even need to make an adjustment.
 
It is a much heavier guage string though, I'm going from a 105 to 125 on the E, I am sure I'll some kind of adjustment. Heavier guage strings tuned lower, will there really that much more tensoin though? Will a 125 tuned to C pull harder then a 105 tuned to E?...
